F17 - Opteon Spectral Analyzer

System Description

The NovoCyte Opteon spectral analyzer features a 5-laser, 73-detector optical configuration that captures the emission spectra of fluorochromes and cell autofluorescence across all five lasers, providing more flexibility regarding fluorophore selection and allows for use of significantly larger panels than in compensation Flow Cytometry. Its dual-laser small particle detection, utilizing both 405 nm and 488 nm side scatter (SSC), allows for the resolution of particles down to approximately 80 nm without requiring manual adjustments between cell and particle modes. To further enhance data clarity, the system supports advanced autofluorescence subtraction by treating autofluorescence as a distinct spectral component, which significantly improves the resolution of dim populations. This entire workflow—from acquisition and unmixing to analysis and reporting—is powered by Agilent’s NovoExpress (Opteon) software version, which includes a dedicated unmixing tab and a streamlined interface for both real-time and offline analysis.
